背景资料	CD28 (Cluster of Differentiation 28) is one of the proteins expressed on T cells that provide co- stimulatory signals required for T cell activation and survival. T cell stimulation through CD28 in addition to the T-cell receptor (TCR) can provide a potent signal for the production of various interleukins (IL-6 in particular). The CD28 gene is mapped to 2q33-q34. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily.

需要用户自己提供的器材和试剂

1.96孔读板仪器。
2.自动洗板机.
3.移液器和枪头。建议使用多通道移液器/排枪。
4.Eppendorf 管。
5.额外洗板缓冲液 (neutral PBS 或 TBS。试剂盒内提供足量。如需额外缓冲液可参考如下配方)。
- 0.01M TBS: Add 1.2g Tris, 8.5g Nacl; 450μl of purified acetic acid or 700μl of concentrated hydrochloric acid to 1000ml H2O and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L.
- 0.01M PBS: Add 8.5g sodium chloride, 1.4g Na2HPO4 and 0.2g NaH2PO4 to 1000ml distilled water and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L.
